Confusing results with GPCR-I-TASSER

Hello,

I am having some difficulty with the results that I got from GPCR-I-TASSER job G00500.

Predicted structures show a structured cytoplasmic domain consisting mainly of alpha helices.
Sequence based predictions show that this region is intrinsically disordered.

I am quite confused about these results. I can share results from sequence based predictions.

HHPred search shows large gaps in this region indicating absence of structural homologue. Region is roughly from 225 to 355.

I looked into the templates used by GPCR-I-TASSER. These templates seem to have same chain ID for two different proteins in the PDB, namely GPCR and soluble cytochrome/lysozyme. I think I-TASSER has taken these non GPCR proteins as GPCR and modelled the target.

To check if the said region is some kind of an insert, I did a BLAST search of it which returned hits for histamine receptors from different species indicating it indeed is part of receptor.

I would like you to have a look into it.

Thank you in appreciation,

Kind regards,

Sagar
