My protein of interest contains two different domains for which structures exist in PDB. I would therefore like to run two different I-TASSER predictions and sort of stitch together the resulting models because I believe that this would increase the predictive value.
My questions are:
1) Is this a useful approach?
2) How is this possible?

I have a PDB-format file for a template structure (one that is not in PDB) that I want to use to build a homology model. It was suggested that I use I-Tasser. I pasted in the sequence I want to model and uploaded the PDB file. However I-Tasser seems to produce results only for sequences already in PDB. How can I get it to use only the uploaded file?

I have a question about how the I-TASSER server uses the template the user specifies. If a template is specified, is this the sole template I-TASSER uses to generate the 5 models? Or does it use this template in addition to other templates found by LOMETS?
